چکیده

We have recently developed a modified ET-Kyoto solution by adding N-acetylcysteine, nitroglycerin, and dibutyryl cyclic adenosine monophosphate to the previously reported ETKyoto solution. The purpose of this study was to compare the new ET-Kyoto solution with the regular Euro-Collins solution and a low-potassium dextran solution containing glucose (Perfadex®) in an isolated rat lung perfusion model. Rat lung blocks were preserved with ET-Kyoto solution, with Euro-Collins solution, or with Perfadex for 4 h. Arterial oxygen tension, shunt fraction, peak inspiratory pressure, mean pulmonary arterial pressure, and pulmonary vascular resistance were assessed up to 50 min after reperfusion. ET-Kyoto solution provided a significantly better lung function after reperfusion than Euro-Collins solution and Perfadex, while Perfadex was also superior to the Euro-Collins solution. We conclude that lung preservation with ET-Kyoto solution is significantly superior to Euro-Collins solution and to Perfadex in an isolated rat lung perfusion model. The tolerable ischemic time, however, is still limited to approximately 7 h [1]. Strategies to increase the ischemic tolerance and as a consequence the supply of transplantable lungs are, therefore, needed. We already reported

Low-potassium dextran solution ameliorates reperfusion injury of the lung and protects surfactant function.

OBJECTIVE This study was designed to compare the effect of lung preservation with low-potassium dextran solution and Euro-Collins solution on reperfusion injury and surfactant function by using an in situ model of warm ischemia. METHODS The left lungs of 6 minipigs were selectively perfused with Euro-Collins solution. In an additional 6 animals low-potassium dextran solution was used for flus...

In vivo measurement of lung preservation solution efficacy: comparison of LPD, UW, EC and low K+-EC following short and extended ischemia.

OBJECTIVE The impact of storage solution composition on graft performance was evaluated following perfusion with either Euro-Collins (EC), low potassium Euro-Collins (rEC), low potassium dextran (LPD) or University of Wisconsin solution (UW) after brief (2 h) and extended ischemia (16 h) in an acute double lung transplantation model in the rat. METHODS Following flush perfusion and ischemia t...

Celsior solution for improvement of currently used clinical standards of lung preservation in an ex vivo rat model.

OBJECTIVE The introduction of Euro-Collins solution with its intracellular electrolyte composition has allowed for clinically accepted pulmonary preservation for up to 7 h of ischemic time. In recent years several alternative solutions have been developed for the improvement of pulmonary preservation. Celsior is an extracellular solution which has significantly reduced the ischemia/reperfusion ...

Impact of lung preservation solutions, Euro-Collins vs. low-potassium dextran, on early graft function: a review of five clinical studies.

Currently, intracellular fluid type solutions that are prepared by modification of Euro-Collins solution (EC) have been the most commonly used by clinical lung transplant programs, which have been associated with a high rate of reperfusion injury. On the basis of the wealth of experimental data demonstrating the benefit of extracellular fluid type solutions over EC, one of the extracellular sol...
